No communication between ALDL or Data master.

For some reason when we are trying to tune this truck we get no communication between the ALDL and the scan tool or data master. Does anybody have any idea why this may be?

What version?
What type of cable? serial or are you using a USB adapter?
What error message?
What message when you press "test com"?

Did you try messing with the ALDL link timing setting in DM? It usually works somewhere between 190 and 200, though some have said theirs will only work at 0. Try EACH option in there, as there may be only one number that will work for you. Disabling the handshake has also made it work for some people.
